So if you think changing the buffer changed the POI, then the simple way to verify this and to have conditions that are truly the same (including environmentals) is to go to the range with both buffers and shoot a target. If correct, then the bullets will group in two different locations roughly 2" apart as you note.
If not satisfied, let somebody else shoot for groups with the 2 buffers.
Otherwise, as has been noted, there is not reason to believe that changing buffers should change your POI.
